My next question is, Does it use requestNumber while running rule for qualification or just only use the rule ?

I think request number is not required because approval rules are triggerred for that specific request number.
Ideally you should add if you have such requirement. Though Rules is triggered as part of Approval Process attached to that request(i.e. through filter triggered with create/modify on particular entry), it will retrieve results from given form based on given qualification. So if you want it to check for specific request number, you should add it to Rule qualification.
